XL10-Gold has the Hte (high transformation efficiency) genotype, which specifically enhances transformation efficiency and the ability to take up large plasmids; it has been successfully used for constructing plasmids up to 40 kb. [Δ(mcrA)183 Δ(mcrCB-hsdSMR-mrr)173] renders XL10-Gold deficient in nearly all known restriction systems. It is also deficient in endonuclease (endA), improving the yield and quality of plasmid DNA. The recA mutation (recombinase deficiency) reduces the probability of homologous recombination of inserted fragments, ensuring the stability of inserted DNA. TetR and CamR confer tetracycline and chloramphenicol resistance. The presence of lacIqZΔM15 enables blue-white screening in XL10-Gold.
