This strain is a lacZY (galactoside permease gene) mutant of the BL21 strain. This mutation allows IPTG to enter each E. coli cell at a uniform rate, resulting in tighter, more homogeneous concentration-dependent induction. It is used for high-level expression of genes cloned into expression vectors containing the T7 promoter (e.g., pET series). The λ phage DE3 region, which contains the T7 RNA polymerase gene, is integrated into the chromosome of Tuner, hence the name Tuner(DE3). It expresses both T7 RNA polymerase and E. coli RNA polymerase and is suitable for protein expression using pET, pGEX, and pMAL plasmids.
