Rosetta 2(DE3) is derived from BL21 and is a derivative of BL21. It is deficient in Lon protease and outer membrane protease OMPT, and the absence of these two proteases effectively prevents degradation of heterologous proteins in E. coli. The introduction of the chloramphenicol- resistant pRARE2 plasmid into BL21(DE3) yields Rosetta 2(DE3). The pRARE2 plasmid supplies tRNAs corresponding to seven rare codons (AUA, AGG, AGA, CUA, CCC, GGA, and CGG) that are deficient in E. coli, enhancing the expression level of exogenous genes. This strain carries the λ phage DE3 region (which contains the T7 RNA polymerase gene) integrated into its chromosome and expresses both T7 RNA polymerase and E. coli RNA polymerase, making it suitable for protein expression using pET, pGEX, and pMAL plasmids.
