This strain is derived from JM109 and is used for high-level expression of genes cloned into expression vectors containing the T7 promoter (e.g., pET series). The λ phage DE3 region, which contains the T7 RNA polymerase gene, is integrated into the chromosome of JM109, hence the name JM109(DE3). It expresses both T7 RNA polymerase and *E. coli* RNA polymerase and is suitable for protein expression using pET, pGEX, and pMAL plasmids. The JM109(DE3) strain cannot be used for blue-white screening.
