HB101 is a hybrid of K12 and B strains (and also the original strain of Stbl3). The recA13 mutation effectively suppresses recombination at terminal repeats of long fragments, reducing the probability of incorrect recombination. The hsdS20 background renders HB101 deficient in the endonuclease system, enhancing the stability and quality of exogenous DNA extraction. This strain exhibits streptomycin resistance. It lacks lacIqZΔM15 and cannot be used for blue-white screening.
