E. coli contains the SbcCD protein complex, which recognizes and excises DNA hairpin structures. Mutations in the sbcC and sbcD genes enhance the stability of DNA hairpin structures. Additionally, introduction of *uidA(DMluI)::pir-116* into the E. coli genome enables GT115 to express the π protein, allowing normal replication of plasmids containing the R6Kγ ori replicon (such as pCpGmcs, pCpG-LacZ, pCpG-siRNA). This strain also carries mutations in endA (nuclease) and recA (recombinase), enhancing the stability of exogenous DNA.
