The EPI400 strain reduces plasmid copy number, making it particularly suitable for cloning unstable DNA or toxic genes. After the addition of WDEPI inducer I, plasmid yield can be restored to normal levels. The [mcrA, Δ(mrr-hsdRMS-mcrBC)] genotype makes the EPI400 strain suitable for cloning DNA rich in methylcytosine or methyladenine. Mutations in recA1 and endA1 facilitate stable insertion of DNA and high-purity plasmid DNA extraction. The presence of the lacZΔM15 marker enables blue-white screening in EPI400. tonA confers resistance to phages T1 and T5, and rpsL confers streptomycin resistance.
