The EPI300 strain contains a mutated trfA gene, whose protein product promotes high-copy amplification of plasmids containing the oriV replicon. The inducer can induce expression of the trfA gene. The [mcrA, Δ(mrr-hsdRMS-mcrBC)] genotype makes the EPI300 strain suitable for cloning DNA rich in methylcytosine or methyladenine. Mutations in recA1 and endA1 facilitate stable insertion of DNA and high-purity plasmid DNA extraction. The presence of the lacZΔM15 marker enables blue-white screening in EPI300, and rpsL confers streptomycin resistance.
