The BL21 Star(DE3) strain is derived from BL21(DE3) and contains the rne131 mutation (RNaseE gene). This mutation reduces the accumulation of endogenous RNase, enhances mRNA stability within the cell, thereby increasing heterologous protein expression levels. It is primarily suitable for high-level protein expression using T7 promoter expression vectors (e.g., pET series) and also contains E. coli RNA polymerase, allowing protein expression with non-T7 promoter expression vectors (e.g., pGEX, pMAL). Due to the relatively high basal expression level of heterologous genes in the BL21 Star(DE3) strain, it is not suitable for expressing toxic proteins.
