BL21(DE3)pLysS integrates the λ phage DE3 region (which contains the T7 phage RNA polymerase gene) and expresses both T7 RNA
polymerase and E. coli RNA polymerase, making it suitable for protein expression using pET, pGEX, and pMAL plasmids. This strain
carries the pLysS plasmid, which confers chloramphenicol resistance and contains the gene encoding T7 lysozyme. T7 lysozyme acts on
the peptidoglycan of the E. coli cell wall, lysing the cells and reducing background expression of the target gene without interfering with
IPTG-induced expression, making it suitable for expressing toxic proteins.
