Exonuclease III is an exonuclease that acts on double-stranded DNA, progressively removing mononucleotides from the 3'-OH terminus. This characteristic allows its use for generating single-stranded DNA in a specific direction. By designing linear DNA with one end as a non-digestible terminus (3' overhang) and the other as a digestible terminus (blunt or 5' overhang), Exonuclease III will digest only one strand.
