T4 DNA Ligase is produced from *E. coli* harboring the T4 phage gene 30. This enzyme repairs single-strand nicks in double-stranded DNA, RNA, or DNA-RNA hybrids, and can ligate DNA fragments with cohesive or blunt ends, but shows no activity on single-stranded nucleic acids. It is primarily used for cloning of restriction enzyme-digested DNA fragments, site-directed mutagenesis, PCR product cloning, linear DNA self-circularization, and repairing nicks in double-stranded DNA. T4 DNA Ligase requires ATP as a cofactor and completes cohesive-end ligation in 10 minutes at room temperature.
