Amplex Red is a peroxidase substrate suitable for absorbance or fluorescence detection. In the presence of peroxidases such as horseradish peroxidase (HRP) or myeloperoxidase, Amplex Red reacts with H₂O₂ in a 1:1 ratio to produce the strongly red-fluorescent compound resorufin. Resorufin has a maximum excitation wavelength of 571 nm and maximum emission wavelength of 585 nm, and also exhibits strong visible light absorption at 570 nm for absorbance detection. This product is sparingly soluble in water, slightly soluble in ethanol (1.5 mg/mL), and soluble in DMSO (~25 mg/mL) and DMF (~25 mg/mL). High-purity Amplex Red can be dissolved in an appropriate buffer solution for direct use in HRP enzyme reactions.
