DAF-FM DA is cell-permeable and, upon entering cells, is cleaved by intracellular esterases to form membrane-impermeable DAF-FM. DAF-FM has weak fluorescence initially but emits strong fluorescence after reacting with nitric oxide, with excitation at 495 nm and emission at 515 nm. Compared to the commonly used NO probe DAF-2 diacetate, DAF-FM DA shows less pH sensitivity, produces more stable fluorescence that resists quenching, and offers higher detection sensitivity with a minimum detection concentration of 3 nM.
