Apoptosis can also be assessed using Hoechst 33342 and propidium iodide (PI) double staining. Hoechst 33342 is a membrane-permeable dye that emits blue fluorescence upon binding to double-stranded DNA. In normal cells, it enters slightly, resulting in low blue fluorescence. Apoptotic cells have increased membrane permeability, allowing more Hoechst 33342 to enter, producing brighter blue fluorescence compared to normal cells. When cells are double-stained and analyzed by flow cytometry or fluorescence microscopy, normal cells appear Hoechst 33342⁺/PI⁺ (low blue/low red), apoptotic cells appear Hoechst 33342⁺⁺/PI⁺ (high blue/low red), and necrotic cells appear Hoechst 33342⁺/PI⁺⁺ (low blue/high red).
