In normal cells, phosphatidylserine (PS) is located on the inner leaflet of the plasma membrane. During early apoptosis, PS translocates to the outer leaflet, becoming exposed on the cell surface. Annexin V has high affinity for PS and is widely used as a sensitive marker for early apoptosis. Propidium iodide (PI) stains cells in late apoptosis or necrosis. Therefore, combining fluorescently labeled Annexin V with PI allows discrimination between different stages of apoptosis.
