Propidium iodide (PI) is a fluorescent dye that binds to double-stranded DNA. Upon binding, the fluorescence intensity is proportional to the DNA content. After PI staining, cells can be analyzed by flow cytometry for DNA content. During early apoptosis, forward light scatter decreases significantly while side scatter increases or remains unchanged. In late apoptosis, both forward and side scatter decrease. Thus, flow cytometry analysis of light scatter changes can assess apoptosis. PI staining is the classic method for cell cycle and apoptosis analysis.
