Cell proliferation assays are fundamental for evaluating cell viability, genotoxicity, and anti-tumor drug efficacy. Our kits include two detection methods: EdU click reaction and CFDA SE staining.
EdU (5-Ethynyl-2'-deoxyuridine) is a thymidine analog containing an ethynyl group. When introduced into animals or cultured cells, it is rapidly incorporated into newly synthesized DNA during cell proliferation. The ethynyl group reacts with fluorescently labeled azide probes (e.g., Alexa Fluor 488, 555, 594, 647) via a copper-catalyzed click reaction, forming a stable triazole ring that labels newly synthesized DNA with fluorescence.
CFDA SE (Carboxyfluorescein diacetate, succinimidyl ester) penetrates cell membranes and is cleaved by intracellular esterases to form CFSE. CFSE covalently and irreversibly binds to lysine residues or other amines on intracellular proteins. Cells are fully labeled approximately 24 hours after CFDA SE addition.
