Calcein AM is derived from Calcein with the addition of an acetoxymethyl (AM) group, which increases hydrophobicity and allows easy penetration through cell membranes. Calcein AM itself is non-fluorescent but produces bright green fluorescence upon hydrolysis by esterases in live cells, whereas dead cells lack this activity. Propidium iodide (PI), a red fluorescent nucleic acid dye, cannot cross intact live cell membranes but enters dead cells to stain the nuclei, producing red fluorescence. Therefore, combined use of Calcein AM and PI enables dual fluorescent staining to distinguish live and dead cells, allowing simultaneous assessment of cell viability and cytotoxicity.
