The Cell Counting Kit-8 (CCK-8) is a rapid, highly sensitive assay kit for cell proliferation and cytotoxicity detection, based on WST-8. In the presence of the electron carrier 1-Methoxy PMS, WST-8 is reduced by mitochondrial dehydrogenases in live cells to form a water-soluble orange-yellow formazan product. The amount of formazan produced is directly proportional to the number of viable cells present. Color intensity increases with cell proliferation and decreases with cytotoxicity, showing a linear relationship with cell number. CCK-8 is widely used in bioactivity factor detection, anti-tumor drug screening, cell proliferation assays, cytotoxicity tests, and drug sensitivity studies.
