During tissue cell isolation, culture, and passaging, cell dissociation reagents are typically used to disperse cells and prepare single-cell suspensions for subsequent subculturing, sampling, and analysis. Commonly used dissociation agents include trypsin, collagenase, and dispase. Trypsin is a serine protease that hydrolyzes intercellular proteins, thereby dissociating cells. Its activity is optimal at pH 8.0 and 37°C. Since different cell types respond differently to trypsin treatment, it is important to control the concentration, exposure time, and temperature to prevent over-digestion and cell damage.
Our trypsin cell digestion solutions are available in various concentrations. The commonly used working concentration is 0.25%. For semi-adherent cells or trypsin-sensitive cells, a concentration of 0.05% can be used directly, or other concentrations can be diluted with PBS to 0.05% to reduce cellular impact. The standard formulation includes EDTA to chelate Ca²⁺ and Mg²⁺, which could otherwise inhibit trypsin activity, ensuring effective cell dissociation. If EDTA may interfere with downstream experiments, an EDTA-free digestion solution is also available.
