In different cell culture experiments, specific nutritional supplements are sometimes added to achieve optimal growth conditions for the cultured cells, or certain components are introduced to influence cells and produce specific effects.
HAT and HT medium supplements mainly consist of hypoxanthine, aminopterin, and thymidine. Under normal conditions, cells have two pathways for purine and pyrimidine synthesis: the de novo synthesis pathway and the salvage pathway. Aminopterin acts as a dihydrofolate analog that blocks the de novo biosynthesis of purines and pyrimidines. Only cells capable of utilizing the salvage pathway can survive. Therefore, HAT/HT medium supplements can be used for the selection of specific cells, such as the selection and validation of hypoxanthine-guanine phosphoribosyltransferase (HPRT)-deficient clones, the production of monoclonal antibodies (mAbs), and luciferase-based bioassays (LUC).

B-27 and N-2 supplements are commonly used to promote the survival and maturation of neurons. During neuronal cell culture, because neurons are highly sensitive and fragile, cell death can easily occur. Extensive research has shown that N-2 supplement supports the growth, differentiation, survival, and expression of post-mitotic neurons in culture. It promotes the expression of neuron-specific genes while inhibiting the growth of non-neuronal cells. B-27 supplement is particularly effective in supporting the long-term maintenance of neurons. L-Glutamine (L-Gln), also known as 2-amino-4-carbamoylbutanoic acid, is the amide of glutamic acid. L-Glutamine is a coding amino acid in protein synthesis and is classified as a non-essential amino acid in mammals, as it can be synthesized from glucose in the body. With a molecular weight of 161.15, it is widely used as a supplement in mammalian cell culture media. L-Glutamine in powder form or frozen solutions is very stable, but it degrades relatively quickly in liquid media or stock solutions. Therefore, fresh L-glutamine supplementation is often required before use to achieve optimal cell performance. This product has a concentration of 200 mM. The solution has been filter-sterilized through a 0.22 μm membrane and is ready for direct use in cell culture and other applications; no additional filtration or sterilization is required before use.

Sodium Pyruvate is a commonly used supplement in cell culture. In addition to glucose, sodium pyruvate can serve as an energy source and is often added to cell culture media as an alternative carbon source. Dexamethasone is a synthetic glucocorticoid, has a molecular weight of 392.5 and CAS number 50-02-2. It is a corticosteroid and an anti-inflammatory glucocorticoid. Dexamethasone has a range of effects on cell survival, cell signal transduction, and gene expression. For example, it inhibits the induction of nitric oxide synthase (IC50 = 5 nM); enhances cation transport in aortic smooth muscle cells by stimulating the Na⁺-K⁺ pump; reduces the activity of cyclin A and Cdk2; inhibits G1/S transition in osteoblasts; suppresses Rb protein phosphorylation; and induces apoptosis in human thymocytes. Lipopolysaccharide (LPS) is a characteristic component of the cell wall of Gram-negative bacteria.
